Design Notes
This year, we decided to improve the ptac parts in the iGEM parts collection by adding ribozymes before the ptac sequence. Such improvement was originally designed for our project to increase the pili yield and now we tried to gather more evidence towards this design. We constructed five plasmid containing ptac and GFP sequence with three different ribozymes addition to them separately: RiboJ, SccJ, SarJ, ptac-new and BBa_K180000. We transformed the plasmid into E.coli BL21 and obtained the data by examining the fluorescence of the GFP produced by E.coli to indicate the effects of different ribozymes with comparison.
